Abstract: This landmark paper introduces the Monod–Wyman–Changeux (MWC) model, providing a structural and thermodynamic framework for understanding allosteric regulation in oligomeric proteins. The authors propose that proteins exist in an equilibrium between distinct conformational states and that ligand binding shifts this equilibrium in a concerted manner. By integrating symmetry considerations with statistical thermodynamics, the model explains the emergence of cooperative, sigmoidal binding behavior without invoking direct site–site interaction. The work establishes a foundational paradigm for interpreting allosteric transitions in biological macromolecules.
